A Novel PCR Method for Detecting ACE Gene Insertion/Deletion Polymorphisms and its Clinical Application

Abstract Background Angiotensin-converting enzyme (ACE) plays a major role in blood pressure regulation and cardiovascular homeostasis. The wide distribution multifunctional properties of ACE suggest it’s involvement various pathophysiological conditions. Results In this study, novel visual detection method for I/D polymorphisms was designed by integrating direct PCR without the need DNA extraction using gold magnetic nanoparticles (GMNPs)-based lateral flow assay (LFA) biosensor. entire procedure could enable genotyping clinical samples about 80 min. limit 0.75 ng results be obtained 5 min LFA device. Three hundred peripheral were analyzed PCR-LFA system then verified sequencing to determine accuracy repeatability. A preliminary study performed analyze total 633 samples. Conclusions After grouping based on age, we found significant difference between genotypes age patients CHD group. introduction into practice may helpful diagnosis diseases caused large fragment gene insertions/deletions.
